Machines (DNA synthesizers) that automate the chemical reactions for the synthesis of DNA have made the production of single-stranded oligonucleotides (<50 nucleotides) a commonplace process. The ability to easily, inexpensively, and rapidly synthesize a strand of DNA with a specified sequence of nucleotides has contributed significantly to the methodologies of molecular cloning and characterization of DNA inserts.
